FOX04-DRI

FOX04-DRI is a D-retro-inverso peptide designed to selectively eliminate senescent cells by disrupting the FOXO4–p53 interaction that normally protects them from apoptosis. Introduced in the 2017 Baar et al. Cell publication as a proof-of-concept senolytic, it has become a workhorse reagent in cellular senescence and aging biology research.

Mechanism of Action

FOXO4 is sharply upregulated in senescent cells where it scaffolds p53 in the nucleus to prevent the apoptotic program. FOX04-DRI competitively displaces p53 from FOXO4, allowing p53 to relocalize to the mitochondria and trigger selective apoptosis of senescent cells while sparing healthy proliferating and quiescent populations.
